Cover: Mps1 protein kinase is a core component of the spindle assembly checkpoint (SAC) signaling, which ensures faithful mitosis. The precise and dynamic kinetochore localization of Mps1 is key to initiate and silence the SAC signaling during prophase and prometaphase, respectively. This work reveals how the Mps1 C-terminal fragment-mediated dimerization contributes to the initial recruitment of Mps1 to the kinetochore, via multisite interactions with Ndc80 complex and enabling responsive SAC signaling.
