Cover: The transcription factor PU.1 serves as a critical regulator of alternatively activated macrophage (AAM) polarization and asthmatic airway inflammation. IL-4-mediated AAM polarization is attenuated in PU.1-deficient macrophages, and partial PU.1-deficient (PU/ER(T)+/−) mice display decreased allergic airway inflammation, which can be restored by adoptive transfer of AAMs. In a triple allergen DRA-induced acute asthma mouse model, the entire left lung is stainedwithH&E (left), and thewhole lung asthmatic inflammation is recorded by an unbiased digital pathology assay (right).
